چکیده

Hepatocellular carcinoma (HCC), the most common primary liver cancer, relies on formation of new blood vessel for growth and frequent intrahepatic extrahepatic metastasis. Therefore, it is important to explore underlying molecular mechanisms tumor angiogenesis HCC. Recently, microRNAs have been shown modulate angiogenic processes by modulating expression critical factors. However, potential roles tumor-derived exosomal in regulating remain be elucidated. In this study, our miRNome sequencing demonstrated that miR-1290 was overexpressed HCC patient serum-derived exosomes, we found delivery into human endothelial cells enhanced their ability. Our results further revealed SMEK1 a direct target cells. MiR-1290 exerted its proangiogenic function, at least part, alleviating inhibition VEGFR2 phosphorylation done SMEK1. Collectively, findings provide evidence promotes via secretion, implicating role as therapeutic

miR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...

Hypoxia-inducible MiR-182 promotes angiogenesis by targeting RASA1 in hepatocellular carcinoma

BACKGROUND Hypoxia is a common feature of solid tumors, including HCC. And hypoxia has been reported to play an important role in HCC progression. However, the potential mechanism of miRNAs in hypoxia mediating HCC progression still remains unclear. METHODS The HCC cells were cultured in the atmosphere of 1 % oxygen to induce hypoxia. The microRNA microarray was employed to search for the hyp...

miR-26b enhances radiosensitivity of hepatocellular carcinoma cells by targeting EphA2

Objective(s): Although low-dose radiotherapy (RT) that involves low collateral damage is more suitable for hepatocellular carcinoma (HCC) than traditional high-dose RT, but to achieve satisfactory therapeutic effect with low-dose RT, it is necessary to sensitize HCC cells to irradiation. This study was aimed to determine whether radiosensitivity of HCC cells can be enhanced using miR-26b by tar...

MicroRNA miR-147b promotes tumor growth via targeting UBE2N in hepatocellular carcinoma

As the subfamily of noncoding RNA, microRNAs (miRNAs) broadly regulate the development of cancers, while their dysregulation and function in human hepatocellular carcinoma (HCC) remains largely unclear. Here, we found the expression level of microRNA-147b (miR-147b) is increased aberrantly in HCC tumor tissues, and its expression positively correlates to the tumor severity. In both MTT and colo...

Downregulation of miRNA-638 promotes angiogenesis and growth of hepatocellular carcinoma by targeting VEGF

The expression and function of microRNA-638 (miR-638) in hepatocellular carcinoma (HCC) remained unknown. Using the miRNA target prediction tools, we predicted that the vascular endothelial growth factor (VEGF) might be a direct target of miR-638. The aim of this study was to test the hypothesis that downregulation of miRNA-638 promotes angiogenesis and growth of HCC by targeting the VEGF signa...
